Vascular and synaptic proteomes reveal blood-brain barrier disruption and postsynaptic remodeling in human temporal lobe epilepsy

Interactive data companion for human MTLE microvessel and PSD-enriched fractions

Summary

This explorer accompanies a study of Microvessels (BBB) and PSD Fraction (Neuronal) isolated from matched epileptic hippocampus and ipsilateral temporal pole tissue from five patients with drug-resistant MTLE.

The data map BBB disruption, vascular inflammation, synaptic remodeling, mitochondrial dysfunction, and neurovascular ligand-receptor changes, with GTEx cross-referencing used to separate disease-associated signals from baseline regional differences.

Proteins are shown as log2 fold-change (hippocampus vs. temporal pole) per patient. Positive values indicate higher abundance in the epileptic hippocampus.

Candidate Compounds

Selected CMap reversal candidates from the vessel, PSD, and combined proteomic signatures. Positive scores indicate predicted reversal of the epileptic hippocampus signature toward the temporal pole state. These are computational candidates, not validated treatments.
Compound Class CMap panel Vessel PSD Combined Network hops
Foretinib MET/VEGFR inhibitor Vessel 4.12 N/A N/A 1.00
Bicalutamide Androgen receptor antagonist Vessel 3.87 N/A N/A 1.00
Sirolimus mTOR inhibitor Combined / Vessel 3.54 N/A 2.91 1.33
Insulin Hormone Vessel 3.21 N/A N/A 1.50
Sorafenib Multi-kinase inhibitor Combined / Vessel 2.98 N/A 2.67 1.82
Isotretinoin Retinoid Combined / PSD / Vessel 2.43 2.11 2.38 2.00
Bexarotene Retinoid Combined / PSD / Vessel 2.19 1.98 2.22 2.00
Anastrozole Aromatase inhibitor PSD N/A 2.54 N/A N/A
Vemurafenib BRAF inhibitor Vessel 2.31 N/A N/A 2.00
Vessel, PSD, and combined columns show CMap reversal scores for each analysis panel. Higher positive values indicate stronger predicted signature reversal. Network hops indicate mean shortest-path proximity from known drug targets to the ECM-integrin module. Lower values indicate closer network proximity. N/A means the compound was not retained as a top candidate in that panel.
